The hydropathy of an amino acid can be used to predict whether an amino acid side chain folds towards the inside or outside of a globular protein. Positive values are hydrophilic while negative values are hydrophobic.

True or false?

Homework Answers

Answer #1

Hydropathy plot is a quantitative representation of hydrophilicity/hydrophobicity of amino acid stretches in a polypeptide chain. It is used to determine the membrane-spanning regions of proteins. Hydrophilic amino acids are located on the surface of a globular protein. Hydrophobic amino acids are buried inside the core of a globular protein.

Hydrophobic amino acids have a positive hydropathy score.
Examples:
Ile = +4.5
Val = +4.2
Phe = +2.8

Hydrophilic amino acids have a negative hydropathy score.
Examples:
Arg = -4.5
Lys = -3.9
Asn = -3.5

So, the given statement is incorrect.

A 500 amino acid protein consists of 200 hydrophobic residues. Upon folding, 150 of the hydrophobic amino acids fold and form a hydrophobic core. Determine the ∆G o for the protein at 300K, considering contributions from both the protein and the water. Assume the following: When the protein folds, each amino acid (hydrophobic and hydrophilic) forms an interaction that is 3 kJ/mol more stable thant the unfolded state. In the unfolded state, the hydrophobic amino acids contact and influence the...

Consider the amino acid histidine, whose side chain pKa = 6.0. Explain what would happen to the histidine pKa if inside of a protein a positively charged lysine was brought into close proximity of the histidine. Do you think the Histidine pKa would increase, decrease, or remain the same? Explain WHY you chose your answer.
