When we do the serial dilution, we can observe that number of turbidity or the number of particles is gradually decreasing and the chances of the number of single-cell colonies are increasing.

When we incubate the Petri dishes ofter pour plating or spread plating of the serially diluted liquid on the gelling media, the number of single colonies of microorganisms should be seen.

Turbidity must be increased after 48 hours if we incubate that culture after 24 hours also and also have the chances of increasing of the number of unrecognized cells.
