For replication of DNA, DNA topoisomerase is essential. These may have been found early on in the recombination as well as in the compaction of chromosome. The most important families are Topo IA, IB, IC, IIA, IIB. These iso,erases are non homologous and this implies that their origination has been independent. Three main topoisomerase had been obtained from eukarya, bacteria, archae. The first toPoisomerase had been known to evolve from gyrase gene. This has been known to come from hyperthermophic origins. This was known to be related evolutionarily due to the presence of the omega protein. These came up by the sign inversion mechanism of gyrase.

Different families of the topo IA, IB as well as IC had come up from different integrates, endonuclease ligases or simply endonucleases, that were involved in the replication of plasmids or viruses.
