In eukaryotes, Primase is a separate RNA polymerase that synthesizes 7-10 nucleotide RNA primer for initiation of DNA replication. The eukaryotic primase is a four-subunit enzyme that contains both RNA primase and DNA polymerase activities in separate subunits, unlike the single subunit bacterial primase. This enzyme is called DNA polymerase alpha-primase. DNA polymerase ? complexes with primase then begins DNA synthesis from RNA primers. pol ?/primase synthesizes RNA primers of 7 -10 nt then extends these another 15 nt with DNA before switching to PCNA and pol ? to finish the job.
