Factors that effect transformation efficiency are:

Bacterial strains: Gram positive bacteria produce a competency factor which makes their neighbours more competent. This results in increasng the transformation efficiency of the entire colony.

Growth phase: Bacteria can be transformed easily during the log or exponential phase. Tranformation efficiency of bacteria is high when it is rapidly dividing.

State of DNA: Supercoiled DNA has a better transformation efficiency than linearized or straight DNA.

Genotype of cells: Tranformation efficiency is greater for cloning strains due to the presence of mutations.

Methods of transformation: Methods that do not use contaminants have a higher rate of transformation.
