After performing the streak plate procedure on a bacterial specimen, the culture was incubated. When you viewed the plate there was heavy growth with no isolated colonies in the first quadrant. There was no other growth on the plate (in second quadrant and beyond). Discuss possible errors that could have led to this result.
Whenever we have to perform streak plate method, then we pick up a colony from the culture of microbial cells and then inoculate it on the nutrient media plate.
In this question there is a heavy growth of bacteria in the first quadrant and almost no growth in the second quadrant. It is because more than one colonies were picked up from the culture of bacteria and inoculated in the first quadrant. All these colonies were transferred to the first quadrant and therefore lead to a heavy growth in this quadrant. But when the needle was moved from 1st to 2nd quadrant, there were no colonies on the needle and therefore no bacteria was seen growing in the second quadrant.
