Mutation or a DNA replication error can cause either deletion or addition of DNA bases which which will lead to to a significant change in the DNA sequence and it will have a a significant effect on the recognition sequence Hence there will be a alteration of recognition site.

Change in the recognition site found on the DNA can we can be analysed by the treating the the DNA whitch is mutated or another DNA which is not mandated with restriction enzymes specific for restriction sites and then run on the gel electrophoresis hence the difference in the number of DNA bands will reveal the change in the restriction sites.
