Deduce what the six wild-type codons may have been in the mutants that led Brenner to infer the nature of the UAG codon

Homework Answers

Answer #1

The codon is UAG which is a stop (Amber) codon as it doesn't code for any amino acid.

To identify six codons, we need 6 amino acids which will be there in the mutant protein. Following amino acids need to be inserted to continue wild-type chain & their codons:

lysine => AAA, AAG

glutamine => CAA, CAG

glutamate => GAA, GAG

tyrosine => UAU, UAC

tryptophan => UGG

SERINE => AGU, AGC, UCU, UCC, UCA, UCG

(In each case, the the Underlined codon will require a single base change to give UAG.)

Conditional alleles allow for mutants to have wild type function in one condition and mutant function in another condition. Temperature-sensitive alleles are one example of conditional alleles that are commonly used. At the permissive temperature, the gene has normal function, whereas at the restrictive temperature, the gene has mutant function. You are studying the Ras signaling pathway and you obtain a temperature-sensitive Ras mutant. At the permissive temperature, Ras functions normally. At the restrictive temperature, Ras is permanently in the...

(True Story) In 1952, the world’s first nuclear reactor meltdown took place at Canada’s Chalk River experimental nuclear station, with a significant release of radioactivity to the surrounding environment. (A U.S. Navy nuclear Engineer named Jimmy Carter participated in the cleanup). As part of a study of the biological effects of the meltdown, microbiologists isolated E.coli strains for analysis. Eventually, one of them was found to have a mutation in the gene encoding a tRNA-tyr. The anticodon of this tyr...

Suppose you have been given responsibility for developing the six-month aggregate production plan at Soda Galore, a manufacturer of soft drinks. Your company makes three types of soft drinks: regular, diet, and super-caffeinated. Fortunately, all three types are made using the same production process, and the costs related to switching between the three types are so minimal that they can be ignored. Thus, you can treat your problem as an aggregate planning exercise where the planning unit is cases of...
