If you wished to design a molecular approach that would distinguish between live and dead bacteria, what would you suggest doing?

Answer #1

When a bacteria is given a nutritional stimulus, the bacteria that is alive will produce a specific rRNA molecule in response to that stimulus but a dead bacteria will not produce it. Hence in live bacteria the levels of rRNA gradually increases whereas in dead bacteria the rRNA levels wont increase at all. This can be identified by designing specific primers for that rRNA and performing a reverse transcriptase-qPCR (RT-qPCR) where we can identify the levels of rRNA gradually increasing in live cells and in dead cells this levels will be constant. This is one of the molecular approach to distinguish between live and dead bacteria.

Halophilic bacteria have been developed to live in a very salty environment, e.g. Dead Sea. The most important salts in the Dead Sea are MgCl2 (1.83 M), NaCl (1.78 M), CaCl2 (0.44 M) and KCl (0.20 M). Typical for halophilic organisms is that their proteins have a much larger (negative) net charge than proteins in organisms that live at normal salinity. (a) Explain why the proteins of halophilic organisms need a large net charge in order for the proteins not...
