1. Sterilize the inoculating loop before placing it in the starter culture. Allow it to cool.

2. Pick up an colony and spread it over the first quadrant back and forth.

3. Flame the loop and allow it to cool again. Go back to edge of the first quadrant and extend the streak towards the second quarter of the plate.

4. Flame the loop again and allow it to cool. Go back to the second area where the last streak was done and start streaking into the third quarter of the plate.

5. Flame the loop again and allow it to cool. Start streaking in the fourth quarter from the edge of third streak.

6. Flame the loop once more.
