mRNA of the polypeptide that has to be transported to ER produces a signal peptide in the N- terminal that comes out of the ribosome during translation. This signal peptide is recognised by a protein called SRC ( Signal recognition complex). SRC bound to signal peptide along with the ribosome binds to the SRC receptor on the surface of the ER situated next to translocon. The signal peptide is inserted in to the ER lumen where it is cleaved by the signal peptidase and protein synthesis is carried on inserting the produced polypeptide in the ER lumen from where it is transported to desired location.
