Phenylalanine hydroxylase enzyme can be purified by single step purification method using affinity methods using its substrate L- phenylalanine. In this method, L- phenylalanine is attached to the agarose matrix and then incubated with the extracted proteins containing the Phenylalanine hydroxylase isolated from rat liver. It has been shown that Phenylalanine hydroxylase binds very tightly to agarose with attached hydrophobic groups (L- phenylalanine). Then the agarose bound Phenylalanine hydroxylase is separated and L- phenylalanine is removed. Removal of L- phenylalanine detaches enzyme from the matrix and thus is purified.
