Staining agents (such as EtBr) usually intercalate between DNA
bases and show up on spectrometers.
As such, the morphology of DNA, whether dispersed as fibres,
condensed as chromatin, assuming the X-like chromatid structure,
these are all possibilities in distribution of cells that are
various stages of the cell cycle.
As such, the nucleus stain appears differently. (Consider the blue
coloured genetic material in the diagram above)
