Explain the steps with DNA sequencing and what happens in each step?
DNA sequencing is used to identify the nucleic acid sequence.
The steps are-
1)- Sample collection or DNA extraction - it is done with the use of enzyme like lipase , protease and other enzymes so that only nucleic acid is left undigested.
2)- PCR amplification - the nucleic acid (DNA) is used to amplify the required genetic material.
3)- impurities purification - such as RNA primer etc
4)- sequencing pre preparation - adapter DNA sequence ligated and amplification is done
5)- DNA sequencing - use of sequencer machine helps in making the sequence of required genetic material .
6)- Daya analysis- the sequence is stored in a computer and matched with normal known DNA and mutation are found . This last step comes under bioinformatics.
