Answer #1

Principle of the experiment : The DNAse agar plate experiment is used to test the ability of an bacteria to produce DNAse.

(Expected) Results :

Positive - When DNA is hydrolysed, methyl green is release which turns the medium colourless around the test bacteria.

Negative - When there is no degradation around the test bacteria i.e the medium remains green.

Conclusion : The colonies producing DNAse hydrolse the DNA. This is evident from the colourless patch aorund these colonies. Whereas, bacteria who dont produce DNAse doesnt hydrolse the DNA.

6 Think about the results you recorded. a. Growth on the MacConkey agar and NA plates was recorded as “good growth,” “poor growth,” or “no growth.” These are qualitative and, at least for the first two, subjective terms. What did you use to establish what constituted “good growth?” b.Why wouldn’t it be advisable to compare growth of the organisms on each plate to each other?
