discuss the process and principle involved for screening/selection of hosts (last stage of cloning) containing the intended recombinant plasmid.

Homework Answers

Answer #1

Hosts are the organisms where the recombinant DNA gets expressed and makes the gene products. The screening and selection of hosts should be done on the basis of-

1. It should easily allow the entry of recombinant plasmid containing desired gene of interest.

2. It should not destroy the recombinant DNA assuming its foreign to the cell, because than the whole idea of gene getting expressed gets nullified.

3. It should have the ability to stably maintain the recombinant DNA, and allow it to replicated inside cell.

4. The transformed host should be easy to handle and maintain.

5. The transformed host should not be able to sustain independently as it can increase the chances of their escape which can prove to be dreadful.

Hope it helps, Good luck !!!!
