If [DNA] determined by DPA reaction and UV measurement is different, which one is more reliable and why?
DNA determination means concentration measurement can be done by either of two methods-
DNA measurement using DPA method involves staining of carbohydrate moity of purine bases by DPA reagent which stains blue. The intensity of colour intensity is now measured at 595 nm the intensity of colour is directly proportional to concentration.
In iv spectrophpotometric method DNA is measured at 230-320nm on spectrophotometer. This method also allows the measurent of common contaminants along with DNA concentration, but the obervation of DNA concentration becomes wrong when contamination level is high.
Thus DNA measurement using DPA reagent is more effective, accurate and reliable method.
