In most of the cases, thiamine deficiency doesn't allow the growth to take place, so thiamine is additionally added. Second reason could be, in mutants, glucose may act as a catabolite repressor in some major pathways. In place of glucose, if sugars like lactose, arabinose or galactose is used with constant shaking then the growth could be observed.
