Briefly, describe the state of the F factor in an Hfr, F+, F', and F- strain of E.coli.

Homework Answers

Answer #1

Answere : State of Hfr, F+, F' and F- strain of E.coli.

F+ cell : In most bacteria, conjugation depends upon fertility factor which is present in F+ cells. It is also called donor cell.

F- cell : The fertility factor is absent in F- cell. It is also called recipient cell.

Hfr cell : In Hfr cell, The fertility factor is integrated inti the bacterial chromosomes. It behave like donor cell forming sex pili and undergo conjugation wiyh F- cell. It is called High frequency cell.

F' cell : In F' cell, the fertility factor present as separate circular DNA, carrying some bacterial genes. It also act as donor cell.

Assume the two E.coli shown below are allowed to conjugate Strain 1: F+, Lac Z, histodine, leucine, ampicillin-resistant Strain 2: F-, Lac Z, histodine -, leucine -, ampicillin-sensitive What is the outcome of conjugation? A) Strain 1 will become F- , LacZ-, histodine-, leucine-, and ampicillin-sensitive; Strain 2 will have an F+, LacZ, histodine+, leucine+, ampicillin-resistant genome B) Strain 1 will remain the same, Strain 2 will have an F+, LacZ, histodine+, leucine+, ampicillin-sensitive genome C) Strain 1 will remain...

Draw a diagram/figure to explain the conjugation process. You should include in the diagram the F- recipient, Hfr Donor and the transconjugant/recombinant recipient. Make sure to include the genes encoding for Leucine, Threonine, Thiamine and Streptomycin resistance in your diagram. How does an Hfr strain of E. coli transfers chromosomal DNA to an F- strain? What determines how much of the chromosomal DNA is transferred?

Derive a resistive deflection bridge suitable for strain measurement using the strain gauge with factor G=2.0, Unrestrained resistance R0=120 Ohm and temperature sensitive to the ambient temperature. The current through the sensor must not exceed 1 mA. Motivate your design choices clearly. Plot the output voltage of the deflection bridge for various choices of the component values in the bridge, illustrating the effect of your design choices.
