What effect will deletion of the trpL region of the trp operon have on the rates of synthesis of the enzymes encoded by the five genes in the trp operon in E. coli cells growing in the presence of tryptophan? Explain your answer.

Answer #1

trrpL is the trp operon leader peptide. This leader peptide has an important role in maintaining or regulating the biosynthesis of tryptophan. In the trpL region the RNA polymerase produces the leader transcript which is translated into the leader peptide - an important part for the tryptophan molecule. Also the trpL region acts as a pause-site for RNA polymerase to make sure that transcription and translation are going on simultaneously. Just after the trpL region is the attenuator sequence. Thus deletion of the trpL region of the trp operon will result to reduction of rates of synthesis of the enzymes encoded by the five genes in the trp operon in E. coli cells and finally stop or attenuate the process in the presence of tryptophan.

Suppose that you used site-specific mutagenesis to modify the trpL sequence such that the two UGG Trp codons at positions 54-56 and 57-59 in the mRNA leader sequence were changed to GGG Gly codons. Will attenuation of the trp operon still be regulated by the presence or absence of tryptophan in the medium in which the E. coli cells are growing? Explain

In the absence of glucose E. coli can metabolize and grow on arabinose, a pentose sugar, using an inducible set of genes that are arranged in three groups on the chromosome (Fig shown). In one of these the araA, araB, andaraD genes encode enzymes for the metabolism of arabinose while the araC gene encodes a regulatory protein that binds adjacent to arabinose promoters and coordinates the expression of the genes in the arabinose operon. The other two groups of genes...
