Proto oncogenes can be transformed into oncogenes by mutations. Some proto oncogenes are involved in cell division. Some in apoptosis. Example of a proto oncogene is RAS.
Tumour suppressor genes suppress the oncogenic potential of the cell. This is by decreasing cell division, repairing errors or inducing apoptosis. Example is P53 gene. When tumour suppressor genes are mutated or removed the cells transform into malignant cells.
In other cases individuals are born with inherited mutations. Example is Familial polyposis coli, Multiple Endocrine Neoplastic Syndrome.
