Poor cell culture technique could result in damage to the cell membrane during the transfer of cells. Why would this type of poor technique be particularly concerning if one was conducting an LDH assay?
Answer:
LDH (Lactate dehydrogenase) assay is the analysis of cell viability. The cytotoxicity is determined by the activity of cytoplasmic enzymes which is released by damaged cells. The damaged cell plays an important role in LDH assay. When the Plasma membrane is damaged LDH is released into cell culture and it is quantified by the NADH produced during the conversion of lactate to Pyruvate.
It also reflects the reduction of Yellow Tetrazolium salt, INT, by NADH into a red, water-soluble formazan. The amount of formazan is directly reflected the amount of LDH in culture and LDH states the no of damaged cells.
