If the concentration of substrate molecules is very high in a reaction medium, then all the enzyme molecules will be bound by substrate molecules. There will be no free enzyme molecule to bind with the leftover substrate molecules.

Kcat is defined as the catalytic efficiency of an enzyme. It is the number of substrate molecules converted into product molecules by one active site of enzyme in a given period of time. It is the ratio of maximum velocity to the total concentration of enzyme in a solution.
