A promoter is the DNA region where RNA polymerase binds to initiate transcription. Promoter includes the entire DNA sequences bound by the enzyme including the transcription start site (+1). RNA synthesis begins from the start site. The transcript is always longer than the coding sequence containing non-coding sequences at the 5' end (leader) upstream of the initiation codon and non-coding trailer (3' UTR) at the 3' end downstream of the termination codon. . Translation of the messenger begins from the start codon in the coding region.
