Azo-proteins are sulfanilamide-azo or sulfanilic acid-azo protein derivatives. They are synthesized by the coupling of diazo compounds. Azo proteins are used during protein synthesis because it was found that in earlier techniques, characterization of proteins with either diazotized arsanilic or sulfanic was highly erroneous since the compounds originally reported to be homogenous mixtures of chromophoric species. These compounds were then successfully purified, characterized by azo proteins. It is also possible to distinguish spectrally between C-2 and C-4 azo coupled histidine.
