The leuk-o tic system consists of 20ul of the blood sample mixed with the 380ul of the buffer solution (that is available in the kit). Hence, the dilution is 1 : 20. This buffer solution lyses only the RBCs leaving the WBCs alive.

The buffer solution also consists of gentian violet stain that stains the WBCs.

The solution is loaded into the haemocyto meter and the cells are contained in the corner large squares.

The formula for counting WBCs is:

WBCs = (Average cell count * Dilution factor)/ (Area (in mm2)* depth ( in mm))

Average cell count = (136+147)/2 = 141.5

Dilution factor = 20

Therefore, WBCs = (141.5 * 20)/ (4 * 0.1) = 7075 = 7.075 * 103 cells / ul

WBCs = 7.075 * 109 cells / L.
