The purpose is to compare the the results that is obtained during the differential test with the control sample.the control sample helps to look into whethere the reagents that are used are contaminated or not . Yes the tube should be incubated with the tubes that are inoculated because controls are always free from actual product undergoing testing so thereby this will confirm the reagents that is used in any particular test is not responsible for any change . And it also ensures that the even if the enviroment conditions changed there wouldnt be any change in the control.
