1) bromophenol blue- it gives blue colour, we mix DNA with loading buffer, the blue colour of the bromophenol -blue helps to identify whether a well is loaded or not, also it gives an idea of how far DNA has migrated through the gel, the bromophenol blue also migrates along the gel.

2) glycerol- glycerol increases the density, so once the DNA is loaded into the well, DNA will not diffuse from the well to the running buffer by the time e load other wells also, glycerol helps the DNA to sink into the well.
