ELISA assay in immunology having several variant, among them two are enzyme linked or based and substrate based. In case of substrate based, which is a type of competitive ELISA, given colored product when used in the experiment can be analyzed through plate reader (chromogenic expression or fluorescence under specific light).

As compared to substrate linked antibodies in ELISA, enzyme linked application is widely used and specific as well as sensitive technique to detect the signal and a type of direct ELISA and enzyme linked antibodies bind to specific antigen (target) precisely as compared to substrate one and substrate linked assay is an indirect application.
