Calibration per ocular unit is calculated as:
Calibration per ocular length = actual length of micrometer/magnification of the lens
In this case first we need to find out the actual length of the micrometer and then use it to calculate the calibration at 400x magnification.
Hence at 20x magnification calibration is 20um. Substituting this data in the above formula we get:
Actual length of micrometer= 20×20 =400um
For an actual length micrometer of 400um and magnification of 400x, the calibration will be:
Calibration = 400/400 = 1 um per ocular unit.
