How p53 can block cancer formation :
p53 is a tumor suppressor protein that can block the cancer formation by inducing DNA repair gene.
When DNA damage occurs in a cell then p53 is accumulated into the cell. It induces DNA repair gene , WAF1. This gene is translated to form p21 protein. This p21 is a regulatory protein that binds with CDK4-CyclinD complex and inactivate them. So the cell is arrested in G1 phase. G1 to S progression is blocked.
P53 also plays a key role in apoptosis that lead to block cancer formation. When excessive DNA damage occurs then p53 does not induce DNA repair gene. It induce the pro apoptotic gene such as Bak, Bax, bid etc. The proteins from such genes help in apoptotic pathway of the cell. Thus the cancer formation is blocked.
