Through the development, approximately one-fifth of the cells in each aggregate differentiated as prestalk cell while the remainder as prespore cells. prestock cell has two subclasses of prestock cell pst A cells and pst O cells. Dif-1 is produced by the prespore cells as part of the negative feedback loop that regulates the pst O to the prespore cell ratio. the ecm A-gene is directly induced by Dif-1 and in the dmtA- strain, mutant that defective in Dif-1 biosynthesis the pstA. specific marker is expressed but the pst-O specific marker is not. these facts suggest that Dif-1 is the physiological inducer of pst-O cell differentiation but not pst-A cell differentiation. by rt-PCR we can detect this expression level.
