Starch digestion bacteria can be identified by using the iodine test for starch on a starch culture plate incubated with the bacterial inoculum of choice. In the starch culture plate, streake of the bacteria of interest are made apart from each other using a sterile swab. The plate is incubated for 48 hours at 37 degrees celcius. When the incubation is complete, a few drops of iodine solution is poured onto the edge of the colonies. The colonies which can digest starch will appear clear, while the rest of the plate will be coloured darkly.
