In amyloclastic method amylase activity is measured by annalysing the decrease in substrate concentration. Sachharogenic method uses the hydrolysis of starch and the amount of reduced sugar formed (or glucose formed ) in a unit time. Hydrolytic method mesures the amount of starch hydrolyzed in a fixed time using the intesity of blue colour produced by the iodine upon reaction with it. Chromolytic method involves measurement of production of soluble dye which is insoluble when associated with starch but soluble in association with simplified or broken down starch.
So Sachharogenic method is the only method which involves measurement of glucose.
