Delbeccos Modified Eagle Media the base media used to culture HeLa cells provides what necessary for proper cell growth?

Homework Answers

Answer #1

DMEM is a basal medium and contains no protein and no growth promoting agents it has almost twice the concentration of amino acids and four times the amount of vitamins.it required suplimentation to be a complete medium.

Original formulation contained 1000m/l of glucose, further variation with 4500mg /l of glucose

It suplimemted with 5_10 % of fetal bovine serum, DMEM utilities a sodium bicarbonate buffer system, because it maintain the required PH,power Media is required by addition of 3.7g/l of sodium bicarbonate upon dissolving in water.

You are given a culture of bacterial cells of unknown concentration. You are asked to determine the concentration (cells/mL). You make three serial dilutions of 100-fold and a final 10-fold dilution of the sample. After plating 1 mL of this culture dilution on a growth plate, you generate 42 bacterial colonies (each colony represents a single bacterial cell that was on the plate). What was the concentration of the original bacterial cell culture?

Therapeutic proteins are expressed in mammalian Chinese Hamster Ovary Cells (CHO), which are grown in rich media broth that often is supplemented with proteins. In this cell culture broth, a receptor containing an Fc group (known as an Fc-fusion protein) was expressed at a titer of 1g receptor/L of cell culture broth. This particular receptor is used to treat Rheumatoid Arthritis by acting as a decoy receptor for Tumor Necrosis Factor alpha and decreasing its binding to its native receptor...

Cells are constantly getting signals that determine whether they survive, grow and divide, differentiate, or die (see figure 16-6). Cells are grown in culture (outside of the body) for a variety of reasons- such as testing drug efficacy and determining the properties of different types of cancers (to name a couple). In order to culture cells, essential nutrients (carbohydrates, amino acids, vitamins, and hormones) need to be present, and these nutrients are provided by using different types of cell culturing...

What is a perfusion culture? What is the main goal? Why is a bleed rate (i.e. the cells removed from the reactor) necessary? Explain the steps needs for downstream purification of a perfusion bioreactor used to produce a monoclonal antibody.

In an experiment, E.coli cells have been genetically modified through site directed mutagenesis. This is a process through which specific locations in an organism’s genome can be altered in order to study the function of such locations, or proteins coded for by genes found in a location. There are many techniques available for site directed mutagenesis. (You have access to PCR ingredients, any primer you wish, laboratory equipment, Lambda phage, and the ability to alter Lambda genome, a commercially available...
