Question

How do PCR and qPCR differ?

How do PCR and qPCR differ?

Homework Answers

Answer #1

Quantitative PCR (qPCR) is used to detect, characterize and quantify nucleic acids.
qPCR (typically green), fluorescent labeling allows the quantification of the amplified DNA molecules
Application - quantification of nucleic acid standards, quantification of next-gen sequencing Libraries and quantification of gene expression.

PCR
PCR, DNA is amplified by 3 repeating steps: denaturation, annealing and elongation.
No fluorescent is use.
No Quantitative analysis
Application - Sequencing, Genotyping, Cloning

Purpose: In this laboratory you will carry out a quantitative real-time PCR (QPCR), using SYBR Green, on the samples that you purified last laboratory session. QPCR serves several purposes in a forensic laboratory and is included in the sample processing workflow for all/most DNA samples. QPCR ultimately measures (either in absolute numbers or relatively) the amount of DNA in a sample and the presence of any inhibitors (either partial or complete). This allows the forensic scientist to determine if more...

4. qPCR using TaqMan probes have the advantage of being very specific for the target species, even when closely related species could also be amplified (e.g., African vs. Indian elephants). TaqMan probes are often 30bp in length. If you had a genome of 3 billion base pairs long: a) provide how many theoretical times that probe should bind to the genome b) explain how the TaqMan probe makes qPCR so much more specific than just having 2 primers to amplify...
