After purifying DNA, an absorbance value at 260nm was found to be 1.50. Based on the calculation method described in your lab manual, what is the concentration in ng/uL?

true or false

Homework Answers

Answer #1

for double-stranded DNA.

The absorbance of 1 at 260 nm corresponds to the concentration of  50µg/ml.

Absorbance is directly proportional to concentration.

A/C= a constant

1/50=constant

so 1.5/x=1/50 ( x is the concentration of DNA sample with absorbance 1.5)

x=1.550

=75 microgram/mL

1 microgram= 10^3 nanogram

and 1 mL= 1000 uL

so 75 microgram/mL= 7510^3 nanogram/1000 microliter=75ng/microliter

so the answer is 75ng/microliter

After converting the nitrate into a purple dye, and measuring the absorbance of the purple dye on a spectrophotometer, a standard curve is used to convert the absorbance into concentration. To make a standard curve, samples with known concentrations of NO3- are run on the spectrophotemeter. The samples with known concentrations are called standards. A linear regression is then performed to relate the concentration of NO3-  to measured absorbance values. Here is a link to a spreadsheet containing a simulated data...

You have a tube of pGLO plasmid at a concentration of 15 ng/uL and you perform transformation experiments using protocol below. The protocols use E. coli that are made competent (i.e. – able to take up exogenous DNA).   Protocol is outlined below: 1 uL of pGLO was added to 100 uL of competent cells in transformation buffer (these cells had been made competent by a different procedure than the one we used in class but we don’t need to concern...

Researchers tested 150 farm-raised salmon for organic contaminants. They found the mean concentration of the carcinogenic insecticide mirex to be 0.0913 parts per million, with a sample standard deviation of 0.0495 ppm. Test, using a 1% level of significance, the claim that the mean concentration of mirex in farm-raised salmon is below the EPAs recommended limit of 0.08 ppm. a. 7) A Value-Added Investment: A “for sale” property has high vacancy. Based on its weak NOI and the seller’s valuation, the property has a cap rate of 3%. Because of the risk (empty space), you are willing to purchase the property at a much higher (above market) going-in cap rate, even though the market has an overall cap rate of 8%. It is your belief that you have found some niche tenants for the property. In the first year, NOI...

This problem is based on information taken from The Merck Manual (a reference manual used in most medical and nursing schools). In the college-age population (18-24 years), about 9.2% have hypertension. Suppose that a blood donor program is taking place in a college dormitory this week (final exams week).
