Integral membrane proteins are those which are completely embedded inside the membrane and span both the surfaces of the membrane. They can be extracted out from the membrane only by disrupting the complete structure of membrane. It is possible by using crude treatment like treating the plasma membrane with detergents. Detergent break down the bonds present between phospholipids in the plasma membrane thereby extracting out integral membrane protein.

Peripheral membrane proteins are those which are present towards the one surface of the membrane. Therefore they can be easily isolated from the membrane without disturbing the structure. This is possible by using mild treatment like high concentration of salt or pH.
