Protein travels from endoplasmic reticulam to Golgi or Golgi to ER through coat protein 2 formed vesicles or coat protein 1 formed vesicles respectively. Hence the protein of interest may be present together with either of the protein

If we tag coat protein 2 with GFP and if it binds with proteins and travel , we can identify through the luminescence and if not found we can also tag the other coat protein 1 with red flourescent protein to identify in which vesicle it travels properly
