If I pirefied DNA by kit for some E.Coli samples , should I send the actual samples with the purified DNA samples to sequence or not ?

Some samples have less than 50 nucleic acid. What should I do with them to get high concentration of nucleic acid ?

Answer #1

Yes u can send the samole as such but only on a condition that they should be processed imidiatly .if the sample will not processed imidiatly then the sample must be refrigerated at a temprature less than 10 degree celcius. U must know that e coli samples must not be held more than 6 hours between collection and initiation of analysis.samples must be processed within 2 hours of reaching the lab

If some.samples has less number of nuclic acid then u can subject those samples to PCR that is polymerase chain reaction which will give u exactly same sample nucleic acid with increase in number .

RNAPpurification. When I was a graduate student, the first protein that I purified was RNA polymerase (RNAP) from E. colicells. RNAP has a net negative charge at physiological pH (7.4), although it binds and transcribes negatively charged DNA. After breaking open cells, I precipated RNAP and other proteins in the cell extract by adding solid (NH4)2SO4to a final concentration of 4 M. I spun down the protein precipitate in a centrifuge and dissolved it in a small amount of water...

DISCUSSION- PRODUCTION THEORY First, the idea that firms (and individuals) should make decisions on how they optimize "on the margin" (where MC=MR). Often people get confused and think about making decisions based on the average (such as average total cost). Average total cost includes FIXED costs, which are sunk and should not influence future decisions. Second, the idea of diminishing marginal returns. That's the idea that each additional unit provides less benefit than the previous unit consumed. Use these concepts...

Some have argued that throwing darts at the stock pages to decide which companies to invest in could be a successful stock-picking strategy. Suppose a researcher decides to test this theory and randomly chooses 100 companies to invest in. After 1 year, 52 of the companies were considered winners; that is, they outperformed other companies in the same investment class. To assess whether the dart-picking strategy resulted in a majority of winners, the researcher tested H 0: P = 0.5...
