A high cAMP concentration increases the transcription of lac operon genes. cAMP is high when glucose is low.

CAP is a positive regulator protein activated by cAMP. The CAP-cAMP complex acts as a regulatory element of lac operon. This complex binds to a specific base sequence called activation sequence in the promoter region in order for transcription of the lac operon to occur. The CAP introduces a >90o bend into DNA at its binding site. This bend in DNA allows CAP to contact RNA polymerase at the promoter and starts transcription from the transcription start site. Thus CAP-cAMP complex is a positive regulator of lac operon.
