anwer) methylation is done by HMT histone methyltransferases and it is added to the lysine and arginine residues of the H3 nad H4 histones. rginine methylationn promotes transcriptional activation while lysine methylation does both trancriptional activation and repression on the methylation site.

now the question is how to identify the new histone methyltransferses for adding post-transcriptional modification on histones

so to identify this kind of modificatiob chIP techinque is used which uses antibodies to isolate the modified proteins along with the DNA sequence. the DNA is mapped to idetify the protein location. if the function of modification is unknown that chIP can be used to infer the function and decoding histone code and helpful in newly discovered modifications. for example : recently identified was ubiqutylation.
