If you discovered a new histone methylation mark, how would you identify the HMT that is responsible for adding this post-translational modification on histones?

Homework Answers

Answer #1

anwer) methylation is done by HMT histone methyltransferases and it is added to the lysine and arginine residues of the H3 nad H4 histones. rginine methylationn promotes transcriptional activation while lysine methylation does both trancriptional activation and repression on the methylation site.

now the question is how to identify the new histone methyltransferses for adding post-transcriptional modification on histones

so to identify this kind of modificatiob chIP techinque is used which uses antibodies to isolate the modified proteins along with the DNA sequence. the DNA is mapped to idetify the protein location. if the function of modification is unknown that chIP can be used to infer the function and decoding histone code and helpful in newly discovered modifications. for example : recently identified was ubiqutylation.

Assume that you have discovered a new mutant phenotype in zebrafish embryos that fails to produce any head structures and is lethal by day 5 of development. Describe a way to rescue the mutant phenotype; i.e., what might you inject to test for this? List some potential genes that when mutated might produce this type of phenotype. Describe how you would identify genes responsible for transcription in E. coli by isolating mutations in their genes. Remember that these genes are required for cell viability. Include all the steps in the isolation of the mutants and also describe how you will differentiate the mutants you want from those in other genes necessary for viability. This includes giving me the expected result for mutants in the genes you want.
