Explanation: During photosynthesis, glucose is formed using light, water & CO2 that is stored as starch molecules in plants.

Third Part:

Note: Assuming this question asked about plants.

Negative iodine test will occur when amount of starch is very low. This will occur when photosynthesis is not occurring in plants & as a result plant will use stored starch. So, two factors that can resulted in negative iodine test are inadequate light & water. These two factors are essential in plants to conduct photosynthesis & absence (or inadequate amount) will hamper photosynthesis.
