The proper autoclaving of samples results in total sterilization of the sample which inactivates every microorganisms including bacteria, viruses and even bacterial spores. Sometimes the improper handling of the media and operating autoclaves may lead to improper sterilization. Some of the reasons behind this are :
- Improper packaging.
- Not providing adequate sterilization time.
- Errors in the functioning of autoclave like gauges not properly working and faulty seals.
- Temperature and time not set properly.
- Not providing adequate spacing between the samples.
All these may lead to improper sterilization which further leads to the presence of bacteria in the autoclaved samples.
