when running the gel electrophoresis Why would the band pattern be different in the tumor DNA than the normal samples?

Homework Answers

Answer #1

DNA obtained from normal patient cells will not have the restriction enzyme site and therefore the amplified DNA will not be cut and will appear as one band. The patient DNA from the tumor sample has a mutation at a particular site and therefore the tumor PCR amplified DNA will be cut to yield two DNA fragments. The peripheral blood and non-tumor patient DNA will have the same pattern where there are three bands due to the combined DNA patterns from the normal gene and the mutant gene.

How could the accuracy and precision of the DNA size estimation using standard gel electrophoresis with Agarose gel be improved? Give 3 ways! P.s. Not too technical perhaps considering it's my first year of university question What I mean is just 3 simple ways to make estimating size of DNA theough gel electrophoresis more accurate or more precise than just manually measuring the distance of DNA bands and comparing it to a reference sample DNA

Imagine that you have just run protein gel electrophoresis using the same protein ladder that we used in this experiment as your molecular weight standard. In the test sample (in the lane just to the right of the ladder), after running the gel you stain for a protein band that is positioned almost exactly halfway between the Phosphorylase and BSA bands on the ladder. A colleague in the lab points to the band and says it must have a molecular...
