Pick a mutation that is caused by either DNA substitution, Insertion or Deletion. (1) What is the error (list the letters) in the DNA?   (2) Did it cause Missense Substitution, Nonsense Substitution or Frame shifting (3) Which Amino Acid or Protein does it create and which amino acid or protein does it normally create (without the mutation)? (3) What effects does it have on the body? (4) List website/references where you retrieved your material.

Answer #1

Normal sequence of DNA,

5' ATG TAC GAA GTG GAC GCA GGG CCC AAT AAA 3'

Normal mRNA sequence,

5' AUG UAC GAA GUG GAC GCA GGG CCC AAU AAA 3'

Amino acid sequence,

N met tyr glu val asp ala gly pro asn lys C

Substitution mutation - replacement of 7th nucleotide G by T

5' ATG TAC TAA GTG GAC GCA GGG CCC AAT AAA 3'

Mutated mRNA sequence,

5' AUG UAC UAA GUG GAC GCA GGG CCC AAU AAA 3'

Mutated amino acid sequence,

5' met tyr stop 3'

This is a type of nonsense mutation because the change in the sequence of nucleotide has lead to the formation of stop codon thereby leading to the premature termination of translation and formation of truncated peptide.
