Describe the synthesis of leading strand during DNA replication. Assume the replication bubbled is open and helicase has already opened the strand.

Homework Answers

Answer #1

Here prokaryotic replication is discussed

As replication fork is already is formed, so process continue with chain elongation.

Chain elongation: DNA chain elongation is catlyzed by enzyme DNA polymerase III using dNTPs (dATP, dCTP,dTTP, dGTP).

Excision of RNA primers and DNA replacement: Excision of RNA primer is carried out by DNA polymerase I by its exonuclease activity (5'->3').

Ligation: Ligation is carried out by enzyme DNA Ligase, it make phosphodiester bond between DNA fragment synthesized and DNA fragments came as replacement of RNA primers.

Termination: Replication halts at Ter site present in organism DNA sequence ( E.coli) with help of tus protein a product of Tus gene. It binds to Ter site and prevents strand displacement.
