Antibiotics are generally used for treating bacteria. Hence, the new antibiotic must be tested for bacterial activity and should not be tested against fungi. For fungi, antifungal agents are used.

To test the efficacy of antibiotics, we have to use Kirby-Bauer agar disc diffusion method. In this technique, a paper disc containing a known concentration of antibiotic will be placed on a lawn of bacteria. The plate will be incubated at respective temperature and after incubation, the zone of inhibition caused by the tested antibiotic will be determined. Based on the diameter of inhibition, the efficacy of antibiotic will be determined.
