Gram staining

Flood the smear with crystal violet and let it stand for 1 minute.

Wash it with tap water.

Then flood the smear with gram iodine mordant.

Wash it with tap water.

Decolorize with 95% ethanol. Add reagent drop by drop until crystal violet fails to wash away from the smear. #Caution - Do not over decolorize.

Wash it with tap water.

Counter stain with safranin for 45 seconds.

Wash it with tap water.

Air dry and examine it under microscope.

Violet colored one is gram positive, and the red one is gram negative.
