protein has signal sequence and the signal sequence is not at the N terminus, once the signal sequence is translated the Signal recognition particle (SRP) binds to it and then SRP binds SRP receptor on the ER membrane and then mRNA-peptide ribosome complex is loaded to the translocon in the ER membrane, the signal sequence will be inserted into the ER lumen.
